Arid1a KO DLL3 CAR-T is an experimental chimeric antigen receptor (CAR) T-cell therapy targeting Delta-like ligand 3 (DLL3), primarily developed for the treatment of small cell lung cancer (SCLC). The therapy incorporates a CRISPR-mediated knockout of Arid1a (a component of the BAF complex) to overcome T-cell exhaustion and limited persistence in the immunosuppressive tumor microenvironment of solid tumors. Research indicates that precise regulation of Arid1a expression enhances the proliferative capacity and stemness of CAR-T cells (marked by TCF1 expression) while reducing the expression of exhaustion markers such as PD-1, TIM-3, and LAG-3. The program includes both autologous and universal (UCAR-T) formats, with studies conducted by researchers at West China Hospital, Sichuan University.
