It is approved for use in schizophrenia, bipolar disorder, and as adjunctive therapy in major depressive disorder. - **Duloxetine** is a serotonin-norepinephrine reuptake inhibitor (SNRI) antidepressant that increases levels of serotonin and norepinephrine by inhibiting their reuptake into neurons. It is indicated for major depressive disorder, generalized anxiety disorder, neuropathic pain, fibromyalgia, and chronic musculoskeletal pain. - **Mirtazapine** is a tetracyclic antidepressant that enhances central noradrenergic and serotonergic activity by antagonizing central presynaptic alpha-2 adrenergic inhibitory autoreceptors and heteroreceptors; it also blocks certain serotonin receptors (notably 5-HT2/3) while acting as an antagonist at histamine H1 receptors. It is mainly used to treat major depressive disorder. - **Nortriptyline** is a tricyclic antidepressant (TCA) that inhibits the reuptake of norepinephrine (and to some extent serotonin), increasing their synaptic concentrations; it also has anticholinergic properties. Nortriptyline treats depression but may be used off-label for neuropathic pain or other indications.
