**ARIZ-047** is a preclinical-stage, ligand-targeted siRNA therapeutic developed by ARIZ Precision Medicine for oncogene-addicted solid tumors, particularly lung cancer. It specifically modulates PRDM2/RIZ gene expression by targeting the RIZ2 isoform to restore the RIZ1/RIZ2 balance, addressing epigenetic dysregulation that drives cell cycle changes and cancer progression. Delivered via a proprietary nanoparticle system with calcium-phosphate formulation and tumor-homing ligands, ARIZ-047 has demonstrated potent efficacy in killing lung cancer cells in vitro and in vivo animal models, outperforming some approved therapies without notable toxicity to normal cells. PRDM2 belongs to the PRDM family of histone methyltransferases implicated in breast, colon, gastric, liver, lung, melanoma, prostate cancers, leukemia, lymphoma, and myeloma.[1][3][5][7]
