ARKD-104 is a novel, brain-penetrant, orally bioavailable small molecule being developed by Arkuda Therapeutics for the treatment of GRN-related frontotemporal dementia (FTD-GRN) and other neurodegenerative disorders. The drug works by increasing progranulin (PGRN) levels in the central nervous system in a dose-dependent manner, addressing the progranulin deficiency caused by heterozygous mutations in the GRN gene. ARKD-104 also increases important cofactors of lysosomal enzymes and enhances the activity of granulins and saposins within the lysosome, thereby restoring lysosomal function. Preclinical studies in cynomolgus monkeys demonstrated that oral administration of ARKD-104 significantly increased progranulin concentrations in the cerebrospinal fluid. The drug is also being explored for the potential treatment of Parkinson's disease and other conditions involving lysosomal dysfunction.
