Arlocabtagene autoleucel is an investigational, autologous CAR T-cell therapy in which a patient's own T lymphocytes are genetically engineered to express a chimeric antigen receptor (CAR) targeting the tumor-associated antigen G-protein coupled receptor family C group 5 member D (GPRC5D). This therapy is designed to recognize and eliminate malignant plasma cells expressing GPRC5D, with potential immunostimulatory and antineoplastic effects. It is being developed primarily for the treatment of relapsed or refractory multiple myeloma, particularly in patients who have received multiple prior lines of therapy. The drug has shown high overall response rates and manageable safety in early-phase clinical trials. Arlocabtagene autoleucel has received regenerative medicine advanced therapy designation from the FDA and is currently being evaluated in phase 2 and phase 3 studies[1][2][3][5][7].
