Overview

ARMed EGFRvIII CAR T is a fourth-generation chimeric antigen receptor (CAR) T-cell therapy engineered to treat glioblastoma (GBM). It specifically targets the tumor-specific epidermal growth factor receptor variant III (EGFRvIII) and is further modified to function as a localized "bio-factory." Upon trafficking to the tumor site, these CAR T cells produce and secrete anti-PD1 mini-antibodies (minibodies) in situ. This dual approach is designed to overcome the highly immunosuppressive tumor microenvironment of GBM and prevent T-cell exhaustion, a limitation observed in earlier generations of EGFRvIII-targeted CAR T therapies. By delivering checkpoint inhibitors directly to the central nervous system, the therapy aims to bypass the limitations of systemic antibody delivery and enhance the durability of the anti-tumor immune response.
