Overview

**Armocibart (KN-057)** is an investigational **monoclonal antibody** developed by **Suzhou Alphamab Biotechnology Co., Ltd.** that inhibits **Tissue Factor Pathway Inhibitor (TFPI)**, a key anticoagulant protein, to promote coagulation and thrombin generation independent of clotting factors VIII or IX. This mechanism addresses unmet needs in **hemophilia A and B**, including patients with inhibitors to factor replacement therapies, by enhancing the extrinsic coagulation pathway via subcutaneous once-weekly administration. It has received **FDA Orphan Drug Designation** for hemophilia A and is in **Phase 3 trials** in China, with Phase 1/2 data showing favorable safety, pharmacokinetics (half-life ~1 week), and pharmacodynamics (increased thrombin potential).[1][2]
