Armored CLDN18.2 nanobody CAR NK cells are an investigational allogeneic cell therapy designed to treat Claudin 18.2-positive solid tumors, particularly gastric cancer. The therapy utilizes Natural Killer (NK) cells derived from cryopreserved cord blood, engineered with a Chimeric Antigen Receptor (CAR) featuring a high-affinity VHH (nanobody) binder specific to the extracellular domain of CLDN18.2. To enhance antitumor activity and persistence in the immunosuppressive solid tumor microenvironment, the construct is armored with an IL-15/IL-15 receptor alpha (sushi domain) complex. This IL-15 signaling promotes the expansion, survival, and metabolic fitness of the NK cells. Developed using a feeder-independent expansion platform, these cells have demonstrated significant regression of gastric cancer xenografts and sustained persistence in preclinical models, supporting future clinical investigation.
