Overview

ARN-75039 is a potent, orally bioavailable small molecule fusion inhibitor targeting a broad spectrum of mammarenaviruses, including both New World and Old World arenaviruses. It acts by inhibiting the envelope glycoprotein complex (GPC)-mediated fusion of viral and host endosomal membranes, thereby blocking viral entry into host cells. Preclinical studies have demonstrated its efficacy in animal models against Tacaribe virus (TCRV) and Junín virus (JUNV), with significant reductions in viral titers and improved survival outcomes when administered prophylactically or therapeutically. ARN-75039 has shown favorable pharmacokinetics with good oral bioavailability and extended half-life in rodents. The drug is under development as a candidate therapeutic for severe arenaviral diseases such as Lassa fever[1][2][3][5][6].
