ARO-ALK7 is an investigational small interfering RNA (siRNA) therapeutic developed by Arrowhead Pharmaceuticals for the treatment of obesity. It is designed to silence the expression of the ACVR1C gene in adipocytes, thereby reducing production of Activin receptor-like kinase 7 (ALK7), a receptor involved in regulating energy homeostasis and fat storage in adipose tissue. By inhibiting ALK7, ARO-ALK7 aims to reduce visceral adiposity and improve lipid and glycemic parameters. Preclinical studies have demonstrated significant reductions in body weight and fat mass with preservation of lean muscle. The drug is currently being evaluated as both a monotherapy and in combination with tirzepatide (a GLP-1/GIP receptor co-agonist) for enhanced effects on weight loss[1][2][3][5][6][8].
