arofylline

Arofylline is a selective phosphodiesterase-4 (PDE4) inhibitor that was developed for the treatment of inflammatory respiratory and skin diseases. By inhibiting the PDE4 enzyme, it increases intracellular levels of cyclic adenosine monophosphate (cAMP), which in turn modulates the activity of various inflammatory cells and reduces the release of pro-inflammatory cytokines. Although it demonstrated efficacy in clinical studies for conditions like asthma, chronic obstructive pulmonary disease (COPD), and canine atopic dermatitis, its clinical utility was limited by significant gastrointestinal adverse effects, such as nausea and vomiting. Development by Almirall was eventually discontinued.
