Overview

Arotinoid acid, also known as TTNPB or Motretinid, is an experimental synthetic retinoid derivative and an analog of retinoic acid. It functions as a potent and selective agonist for retinoic acid receptors (RARs), specifically RARα, RARβ, and RARγ. By binding to these nuclear receptors, which are transcription factors, it modulates gene expression involved in cellular differentiation, proliferation, and apoptosis. While initially developed for potential medical applications in cancer treatment and dermatological conditions such as psoriasis and acne, its clinical adoption was limited due to its potent teratogenic activity. More recently, Arotinoid acid has found utility in research as part of chemical cocktails for cellular reprogramming to produce pluripotent stem cells. In cellular models, it has been shown to inhibit cell growth, induce cell cycle arrest in the G0/G1 phase, and promote apoptosis, primarily by suppressing the activity of Cyclin D1 and Cyclin D3 proteins.
