Overview

ARRY-403 (also known as AMG-151) is a potent, selective, orally administered small molecule glucokinase activator developed for the treatment of type 2 diabetes. It works by stimulating glucokinase activity in both the pancreas and liver. In the pancreas, this enhances glucose sensing and insulin secretion in response to elevated blood glucose levels; in the liver, it increases glucose utilization and decreases hepatic glucose production. Clinical studies demonstrated that ARRY-403 reduced both fasting and postprandial blood glucose levels in patients with type 2 diabetes inadequately controlled with metformin. The drug was originally discovered by Array BioPharma and later developed through a partnership with Amgen. Despite promising early results, development of ARRY-403 for type 2 diabetes was discontinued[1][2][3][5][6].
