Overview

ARSB siRNA is a small interfering RNA (siRNA) designed to silence the expression of the enzyme arylsulfatase B (ARSB), also known as N-acetylgalactosamine 4-sulfatase. ARSB is a lysosomal enzyme responsible for the degradation of sulfated glycosaminoglycans, specifically by removing 4-sulfate groups from N-acetylgalactosamine 4-sulfate residues in chondroitin 4-sulfate (C4S) and dermatan sulfate (DS). In research contexts, particularly in melanoma, silencing ARSB leads to an accumulation of highly sulfated C4S, which triggers transcriptional pathways involving galectin-3, AP-1, and Sp1. This results in the upregulation of chondroitin sulfate proteoglycan 4 (CSPG4) and increased matrix metalloproteinase (MMP) activity, thereby enhancing the invasive potential of melanoma cells. It is primarily utilized as a laboratory tool to investigate the role of ARSB in cancer progression and glycosaminoglycan metabolism.
