Overview

Arsenic hexoxide (As4O6) is an inorganic polymorphic arsenic compound developed by Chemas as an anticancer therapeutic. Originally derived from the natural mineral Sinsuk through a specialized purification process designed to eliminate toxicity, arsenic hexoxide exhibits potent antitumor activity across various malignancies, including cervical, bladder, brain, breast, colon, and non-small cell lung cancers. Its mechanism of action involves the induction of programmed cell death, G2/M cell cycle arrest, and autophagy. Specifically, arsenic hexoxide inhibits STAT3 phosphorylation, leading to mitochondrial reactive oxygen species (ROS) generation and caspase-3-dependent cleavage of gasdermin E (GSDME), which triggers pyroptosis in triple-negative breast cancer (TNBC) cells. It also suppresses the activation of Akt and VEGFR2, and downregulates NF-κB activity, thereby inhibiting angiogenesis, cell proliferation, and metastasis.
