Overview

ART-838 is a synthetic, artemisinin-derived trioxane diphenylphosphate dimer developed for the treatment of acute leukemias, including acute myeloid leukemia (AML). It is a more potent derivative of artesunate, a semi-synthetic artemisinin used for malaria. ART-838 was developed through a collaboration between researchers at Johns Hopkins University, who synthesized the compound, and the University of Maryland, who evaluated its biological activity. The drug exhibits broad antineoplastic activity by inhibiting the growth of leukemia cells and inducing apoptosis while sparing normal CD34+ hematopoietic stem and progenitor cells. ART-838 has demonstrated significant synergy when combined with BCL2 inhibitors, such as venetoclax (ABT-199), and other antileukemic agents in both in vitro and in vivo models.
