ART-I02 is a **recombinant adeno-associated virus serotype 5 (rAAV5) gene therapy** vector encoding **human interferon-beta (hIFN-β)** under the control of a nuclear factor κB (NF-κB)-responsive promoter[1][5][3][2]. The NF-κB promoter enables transgene expression primarily in inflamed tissue, such as arthritic joints. ART-I02 is designed for **local intra-articular administration** to induce sustained, regulated production of anti-inflammatory human IFN-β in synovial cells, especially fibroblast-like synoviocytes. Its primary indication is for the treatment of **rheumatoid arthritis** (RA), with application also considered for **inflammatory hand arthritis** (IHA). Preclinical studies have shown potent, localized bioactivity and favorable biodistribution, with vector remaining largely in injected joints. Initial clinical data indicate local moderate adverse events leading to discontinued recruitment, but no serious systemic safety concerns. ART-I02 is being developed as a local therapy for patients with persistent joint inflammation unresponsive to standard systemic treatments[1][3][5][8].
