Overview

Artefenomel is a novel synthetic trioxolane antimalarial small molecule, also known as OZ439. It is designed to improve upon the pharmacokinetic properties of artemisinin derivatives, offering a longer half-life and the potential for single-dose cure in combination therapies. Artefenomel acts rapidly against the asexual blood stages of Plasmodium falciparum and Plasmodium vivax malaria, with demonstrated transmission-blocking activity. Its mechanism involves reductive activation by heme within the parasite, generating radicals that alkylate proteins or other biological targets; however, its precise molecular target remains unknown. The drug has been generally well tolerated in clinical trials up to phase II and is being developed primarily as part of combination regimens for uncomplicated malaria[1][2][6][8].
