Artemether + lumefantrine + amodiaquine is a fixed-dose triple combination antimalarial therapy used for the treatment of uncomplicated Plasmodium falciparum malaria, particularly in regions with multidrug-resistant parasites. This regimen combines three agents: - Artemether, a fast-acting artemisinin derivative that rapidly reduces parasite burden. - Lumefantrine, a long-acting partner drug that helps prevent recrudescence. - Amodiaquine, another long-acting antimalarial with a different mechanism of action. The combination aims to improve cure rates and delay the development and spread of drug resistance by attacking the malaria parasite through multiple mechanisms. Clinical studies have shown this triple therapy to be well tolerated and highly effective in reducing both clinical symptoms and transmission potential within 48 hours after initiation[1][2][5]. The addition of amodiaquine to standard artemether-lumefantrine approximately halves the risk of recrudescence compared to dual therapy[1]. Side effects are generally mild but may include increased incidence of vomiting or mild bradycardia[1][2].
