When co-administered, efavirenz (a CYP3A4 inducer) significantly reduces plasma concentrations of artemether, its active metabolite dihydroartemisinin (DHA), and lumefantrine, potentially compromising antimalarial efficacy. Day 7 lumefantrine levels, considered predictive of treatment success, are reduced by approximately 46% with efavirenz co-administration.
