Overview

This entry refers to a combination of four antimalarial agents: artemether, lumefantrine, sulfadoxine, and pyrimethamine. In the context of clinical research by the Institute of Tropical Medicine, Belgium, these agents were evaluated as two distinct fixed-dose combinations—artemether-lumefantrine (marketed as Coartem) and sulfadoxine-pyrimethamine (marketed as Fansidar or SP)—to compare their safety and efficacy in treating uncomplicated *Plasmodium falciparum* malaria in Zambia, particularly among HIV-infected and non-infected adults. Artemether and lumefantrine act by generating reactive free radicals and inhibiting hemozoin formation, respectively, providing rapid parasite clearance and long-acting suppression. Sulfadoxine and pyrimethamine act synergistically to inhibit sequential steps in the parasite's folic acid synthesis pathway, targeting dihydropteroate synthase (DHPS) and dihydrofolate reductase (DHFR).
