Artificial tears preserved with benzalkonium chloride (AT-BAK) are ophthalmic solutions evaluated by Mati Therapeutics as an adjunctive penetration enhancer for the Latanoprost Punctal Plug Delivery System (L-PPDS; Evolute). Benzalkonium chloride (BAK) is a quaternary ammonium surfactant that temporarily disrupts the tight junctions and lipid bilayer of the corneal epithelium. This disruption increases the permeability of the cornea, thereby facilitating the intraocular absorption of latanoprost released from the punctal plug into the tear film. The combination approach is intended to augment the intraocular pressure (IOP) lowering efficacy of the punctal plug system in patients with open-angle glaucoma or ocular hypertension. This strategy was investigated in a Phase 2 clinical trial to optimize drug delivery and therapeutic response.
