Overview

artLCMV-TRP2 is a replicating but attenuated arenavirus-based therapeutic cancer vaccine. The vector is derived from lymphocytic choriomeningitis virus (LCMV) and has been engineered to carry the gene for the melanoma-associated self-antigen tyrosinase-related protein 2 (TRP2). This allows the vaccine to induce robust TRP2-specific CD8+ T cell responses, effectively breaking immune tolerance to the self-antigen and promoting cytotoxic T cell infiltration and function within melanoma tumors. The vector’s replication and antigen persistence are essential for efficient antigen presentation and strong activation of professional antigen-presenting cells (APCs), particularly dendritic cells and macrophages. It is indicated for preclinical and translational studies in melanoma, primarily in mouse models, and has demonstrated the ability to inhibit tumor growth, enhance survival, and provide long-term protection against melanoma recurrence. The vaccine is also being explored in combination with immunotherapies such as adoptive T cell transfer[1][2][4][6][7].
