ARTS-011 is a highly selective, potent, and orally available small molecule inhibitor of Tyrosine kinase 2 (TYK2), developed as a potential therapeutic for autoimmune and inflammatory diseases. It acts as an allosteric inhibitor by binding to the JH2 regulatory domain of TYK2, thereby blocking signal transduction downstream of cytokines such as interferons (IFN), interleukin-12 (IL-12), and interleukin-23 (IL-23). These pathways are implicated in the pathogenesis of conditions like psoriasis, lupus, and inflammatory bowel disease. Preclinical studies have demonstrated that ARTS-011 effectively inhibits relevant signaling pathways in cellular assays and animal models for psoriasis and IBD. The drug is currently being evaluated in early-phase clinical trials for autoimmune diseases including psoriasis.
