Drug intelligence / Profile preview
ARTS mimetic peptides are synthetic peptides designed to mimic the pro-apoptotic function of the mitochondrial tumor suppressor protein ARTS (also known as Sept4_i2). These peptides are derived from the unique C-terminal domain of ARTS, specifically the ARTS-IAP-binding motif (AIBM). They act by directly binding to the BIR3 domain of X-linked inhibitor of apoptosis protein (XIAP), promoting its auto-ubiquitylation and subsequent degradation via the ubiquitin-proteasome system (UPS). This degradation relieves the inhibition of caspases (such as caspase-3 and caspase-9), thereby triggering apoptotic cell death in cancer cells. Research has demonstrated their potential in treating various cancers, including glioblastoma, where they have been evaluated in combination with autophagy inhibitors to overcome apoptosis resistance.
