Overview

ARV-6723 is an investigational, orally bioavailable PROTAC (PROteolysis TArgeting Chimera) designed to selectively degrade Hematopoietic Progenitor Kinase 1 (HPK1). Developed by Arvinas, it is the company's first clinical candidate in the immuno-oncology (IO) space. HPK1 is a kinase primarily expressed in hematopoietic cells that acts as a negative regulator of T-cell receptor signaling; by degrading both the kinase and scaffolding functions of HPK1, ARV-6723 aims to enhance the immune system's ability to mount a potent anti-tumor response. Preclinical studies have demonstrated selective HPK1 degradation and superior tumor control in both low- and high-immunogenic tumor models. Arvinas plans to initiate a Phase 1 clinical trial for ARV-6723 in patients with advanced solid tumors in 2026.
