Drug intelligence / Profile preview
ARVib-7 is a novel, orally bioavailable small molecule niclosamide analog developed by researchers at the University of California, Davis and The Ohio State University. It was designed to overcome the poor bioavailability and potency limitations of its parent compound, niclosamide. ARVib-7 acts as a potent inhibitor of NADH:ubiquinone oxidoreductase subunit S4 (NDUFS4), a nuclear-encoded subunit of mitochondrial electron transport chain complex I. By targeting NDUFS4, ARVib-7 disrupts oxidative phosphorylation, suppresses mitochondrial respiration, and induces ferroptotic stress (characterized by intracellular iron accumulation and altered expression of markers like COX2, CHAC1, NRF2, and GPX4). Additionally, ARVib-7 functions as an androgen receptor (AR) and AR splice variant 7 (AR-V7) degrader by suppressing HSP70 expression and promoting the nuclear translocation of the E3 ubiquitin ligase STUB1, which facilitates the ubiquitination and proteasomal degradation of AR/AR-V7. ARVib-7 is being investigated preclinically for the treatment of advanced, castration-resistant, and therapy-resistant prostate cancers, including those resistant to androgen receptor signaling inhibitors (such as enzalutamide and abiraterone) and PARP inhibitors (such as olaparib).
