Overview

ARX102 is a precision-engineered, modified interleukin-2 (IL-2) biologic designed for immuno-oncology applications. It is a PEGylated IL-2 molecule expressed in mammalian cells with glycosylation, which confers full functionality similar to native human IL-2. The drug has been engineered to drastically reduce binding to the IL-2 receptor alpha subunit (IL2Rα), while selectively targeting the beta and gamma subunits on cytotoxic T cells. This selectivity aims to stimulate anti-tumor immune responses by increasing intratumoral CD8+ T cell infiltration and activity, while minimizing off-target effects associated with traditional IL-2 therapies. Preclinical studies have demonstrated potent anti-tumor activity as a single agent in models of breast cancer, colorectal cancer, melanoma, and renal cell carcinoma[5][7]. ARX102 was initially developed by Ambrx and is being co-developed with Sino Biopharm for clinical advancement[3][5].
