Overview

Aryloboronate doxorubicin prodrugs are a class of reactive oxygen species (ROS)-responsive small molecule anticancer agents. These prodrugs consist of the chemotherapy agent doxorubicin conjugated to an arylboronate moiety, which serves as a trigger for selective drug release in the high-ROS environment characteristic of cancer cells. Preclinical studies, particularly in pancreatic cancer models like MiaPaCa-2, have demonstrated that these compounds can effectively release free doxorubicin within tumors, leading to tumor regression while potentially reducing systemic toxicity compared to the parent drug. The benzeneboronate derivative has shown significant cytotoxicity and tumor regression in ovo models.
