Overview

AS-19 is a potent and selective small molecule agonist of the 5-HT7 receptor, characterized by a high affinity (Ki approximately 0.6-0.8 nM). It is primarily utilized as a pharmacological research tool to investigate the physiological and pathological roles of the 5-HT7 receptor in the central nervous system. Preclinical studies have demonstrated that AS-19 can enhance memory consolidation and exert antinociceptive effects in various animal models, including those for chronic pain and knee osteoarthritis. Its mechanism of action involves the activation of the Gs-protein-coupled 5-HT7 receptor, which stimulates the adenylyl cyclase-cAMP-PKA signaling pathway. In the spinal cord, this pathway is linked to the activation of GABAergic inhibitory neurons, which contributes to the modulation of pain transmission. While it has shown promise in research for cognitive enhancement and analgesia, it is not currently approved for clinical use in humans.
