AS-5 is a synthetic small molecule derivative of resveratrol that acts as a potent activator of Sirtuin 1 (SIRT1). SIRT1 is a nicotinamide adenine dinucleotide (NAD+)-dependent deacetylase that plays a critical role in regulating cellular metabolism, stress responses, and aging processes. By activating SIRT1, AS-5 mimics the beneficial effects of calorie restriction, potentially offering therapeutic benefits for metabolic disorders, neurodegenerative diseases, and age-related conditions. Preclinical studies have demonstrated that AS-5 can improve mitochondrial function, enhance insulin sensitivity, and provide neuroprotection in various models of disease. While it has been widely utilized as a pharmacological tool in academic research to investigate SIRT1 biology, its clinical development as a pharmaceutical drug has been limited compared to other sirtuin-activating compounds (STACs).
