Drug intelligence / Profile preview
**AS01B** is GSK's proprietary liposome-based vaccine adjuvant system rather than a standalone antigen-containing vaccine. It combines the immunostimulants 3-O-desacyl-4'-monophosphoryl lipid A (MPL), a detoxified *Salmonella minnesota* lipopolysaccharide derivative and Toll-like receptor 4 agonist, with the *Quillaja saponaria*-derived saponin QS-21 in DOPC and cholesterol liposomes. The formulation promotes transient innate immune activation, antigen-presenting-cell recruitment and activation, and enhanced antigen-specific cellular and humoral immunity. It is a component of the recombinant zoster vaccine Shingrix and has also been evaluated as an adjuvant in investigational infectious-disease vaccines and intratumoral cancer-immunotherapy regimens.
