The AS01E adjuvant system is a liposome-based immunological adjuvant developed by GSK, specifically utilized as a control or comparator in clinical trials for tuberculosis vaccine candidates like M72/AS01E (also known as GSK 692342). It is composed of two key immunostimulants: 3-O-desacyl-4'-monophosphoryl lipid A (MPL) and the saponin QS-21, formulated within a liposomal carrier. MPL is a detoxified derivative of lipopolysaccharide that acts as a Toll-like receptor 4 (TLR4) agonist, while QS-21 is a triterpene glycoside extracted from the bark of the Quillaja saponaria tree that activates the NLRP3 inflammasome. Together, these components synergistically enhance the magnitude and quality of the cellular and humoral immune responses. In the context of tuberculosis research, this adjuvant-only formulation serves as a critical control to differentiate the safety, reactogenicity, and background immunogenicity of the adjuvant system from the effects of the recombinant tuberculosis antigens.
