AS03-adjuvanted influenza vaccine

AS03-adjuvanted influenza vaccine (AS03-TIV) is an inactivated trivalent split-virion seasonal influenza vaccine developed by GSK. It combines influenza hemagglutinin (HA) antigens from recommended seasonal strains with the proprietary AS03 Adjuvant System, an oil-in-water emulsion containing α-tocopherol, squalene, and polysorbate-80. The adjuvant is specifically designed to enhance the immunogenicity of the influenza antigens, particularly in elderly populations (≥60 years) where immune responses to standard, non-adjuvanted vaccines are often suboptimal due to immunosenescence. The vaccine is administered via intramuscular injection and aims to stimulate both humoral and cellular immune responses to prevent seasonal influenza infection. Clinical evaluation has included Phase IIb trials (such as FLU-LD-002) assessing the safety and immunogenicity of low-dose formulations compared to standard non-adjuvanted vaccines like Fluarix.
