AS03-adjuvanted low-dose influenza vaccine

The AS03-adjuvanted low-dose influenza vaccine is an investigational seasonal influenza vaccine developed by GSK Biologicals. It is a trivalent, inactivated split-virion vaccine based on the Fluarix platform but formulated with a significantly reduced antigen content of 5 μg of hemagglutinin (HA) per strain (A/H1N1, A/H3N2, and B), compared to the standard 15 μg. The vaccine incorporates GSK's proprietary AS03 Adjuvant System, an oil-in-water emulsion containing squalene, α-tocopherol, and polysorbate 80. This adjuvant is designed to enhance the magnitude and breadth of the immune response by stimulating antigen-specific memory B cells and polyfunctional CD4+ T cells. The vaccine has been primarily evaluated in Phase 2 clinical trials, particularly in older adults (aged 60 and older), to address the age-related decline in vaccine efficacy (immunosenescence) by providing a more robust and cross-reactive antibody response.
