The AS03-adjuvanted low-dose trivalent influenza vaccine is an investigational inactivated split-virion vaccine developed by GSK for the prevention of seasonal influenza, particularly in older adults. The formulation combines a reduced dose of hemagglutinin (HA) antigens from three influenza strains (typically A/H1N1, A/H3N2, and a B strain) with the AS03 Adjuvant System. AS03 is an oil-in-water emulsion consisting of squalene, alpha-tocopherol (α-tocopherol), and polysorbate-80, designed to enhance the magnitude and breadth of the immune response. This adjuvant-mediated enhancement allows for antigen dose-sparing while maintaining or improving immunogenicity compared to standard unadjuvanted vaccines. The vaccine primarily targets the viral hemagglutinin protein to stimulate hemagglutination-inhibition (HI) antibody responses and CD4+ T-cell mediated immunity.
