Drug intelligence / Profile preview
AS101-003 is a next-generation, first-in-class organotellurium small molecule antibiotic developed by researchers at Kaohsiung Medical University and MacKay Medical College. Derived from the clinical-stage hypervalent tellurium compound AS101, AS101-003 is designed to target carbapenem-resistant Gram-negative bacteria, including Acinetobacter baumannii, Klebsiella pneumoniae, and Escherichia coli, as well as Neisseria gonorrhoeae and Elizabethkingia meningoseptica. It exerts its antibacterial activity by disrupting bacterial membrane integrity and interfering with glutathione metabolism, which induces the accumulation of reactive oxygen species (ROS) and leads to rapid bacterial cell death. AS101-003 has demonstrated potent in vitro and in vivo efficacy in mouse sepsis and vaginal infection models, showing a low frequency of spontaneous resistance and improved safety compared to its parent compound AS101.
