Overview

**AS1266** is a small molecule allosteric tyrosine kinase inhibitor (TKI) in preclinical development targeting the BCR::ABL fusion protein, specifically binding to the myristoyl pocket on the ABL kinase domain. This STAMP (Specifically Targeting the ABL Myristoyl Pocket) mechanism induces a conformational change that inhibits downstream signaling, offering potential efficacy against chronic myeloid leukemia (CML), including cases resistant to conventional ATP-competitive TKIs or harboring the T315I gatekeeper mutation.[3][4][5]
