Overview

AS1409 is a genetically engineered fusion protein designed as an immunocytokine for cancer therapy. It consists of a humanized monoclonal antibody (BC1) that targets the ED-B variant of fibronectin, which is associated with tumor vasculature, fused to the cytokine interleukin-12 (IL-12). The mechanism of action involves targeted delivery of IL-12 to tumor-associated blood vessels, aiming to stimulate anti-tumor immune responses while minimizing systemic toxicity. Preclinical studies demonstrated enhanced anti-cancer activity compared to untargeted IL-12. In phase 1 clinical trials, AS1409 was investigated for renal cell carcinoma and malignant melanoma, showing evidence of immune activation and some clinical benefit at the maximum tolerated dose[1][2][3][4][5][7].
