AS1411 + let-7 is an experimental targeted oligonucleotide delivery system designed for the treatment of various cancers, most notably breast and gastric cancers. The system utilizes the AS1411 DNA aptamer as a targeting ligand that specifically binds to nucleolin, a protein frequently overexpressed on the plasma membrane of malignant cells. This aptamer is typically conjugated to a delivery vehicle, such as extracellular vesicles (EVs) or nanoparticles, which encapsulate the let-7 microRNA (miRNA). Upon binding and internalization into the tumor cell, the let-7 miRNA is released to exert its tumor-suppressive effects by downregulating key oncogenes, including RAS, c-MYC, and STAT3. This dual-component approach aims to enhance the therapeutic index of miRNA therapy by ensuring selective delivery to tumor tissues while reducing off-target effects in healthy cells.
