AS15 is a vaccine adjuvant system developed to enhance the immune response to antigens in cancer vaccines. It contains CpG 7909 (a synthetic oligodeoxynucleotide that acts as a TLR9 agonist), monophosphoryl lipid A (a detoxified derivative of lipopolysaccharide acting as a TLR4 agonist), and QS-21 (a saponin immunostimulant). AS15 has been used in combination with recombinant tumor antigens such as recMAGE-A3 for antigen-specific immunotherapy in oncology, including prostate cancer and multiple myeloma. Its mechanism involves stimulating both innate and adaptive immunity, leading to enhanced antigen presentation and T-cell activation[2][3][5].
