Overview

AS1517499 is a **small molecule** selective inhibitor of **signal transducer and activator of transcription 6 (STAT6)**, originally developed by **Astellas Pharma**. It blocks the STAT6 signaling pathway which is critical for Th2-mediated immune responses, particularly downstream of cytokines interleukin-4 (IL-4) and interleukin-13 (IL-13). Preclinical studies have demonstrated that AS1517499 reduces Th2-related cytokine levels, alleviates airway inflammation, and suppresses features of atopic dermatitis and asthma in animal models by specifically blocking STAT6. The drug has also been shown to reduce the upregulation of RhoA and IL-13 production, which are linked to bronchial hyperresponsiveness, and may have potential for treatment of allergic and Th2-mediated diseases such as asthma and atopic dermatitis. The compound is a derivative of 2-{\[2-(4-hydroxyphenyl)ethyl\]amino}pyrimidine-5-carboxamide.
