AS2762900-00 is a fully human monoclonal antibody that specifically targets the interleukin-23 receptor (IL-23R). Developed by Astellas Pharma, it was designed for the treatment of chronic inflammatory disorders, including psoriasis and inflammatory bowel disease (IBD). Unlike many other biologics in this space that target the IL-23 cytokine (such as the p19 or p40 subunits), AS2762900-00 binds directly to the receptor itself. This approach is hypothesized to be more efficient because receptor expression levels are typically lower than circulating cytokine levels, potentially allowing for more complete blockade of the IL-23/IL-17 signaling axis. In preclinical studies, the antibody demonstrated the ability to inhibit IL-23-induced cell proliferation and reduce systemic Th1 and Th17 inflammatory responses. Despite its mechanistic promise, clinical development of the compound was discontinued.
