Overview

Asaretoclax (ZN-d5) is an orally bioavailable, selective small molecule inhibitor of B-cell lymphoma 2 (BCL-2), an anti-apoptotic protein frequently overexpressed in various hematologic malignancies. By binding to BCL-2, asaretoclax restores the apoptotic signaling pathway, leading to the programmed cell death of cancer cells. It is being developed by Zentera Therapeutics (a joint venture between Zentalis Pharmaceuticals and a consortium of investors) primarily for the treatment of B-cell malignancies, including non-Hodgkin lymphoma (NHL) and potentially acute myeloid leukemia (AML). Clinical development is currently focused on the Chinese market, where it is being evaluated in Phase I studies for safety, pharmacokinetics, and clinical activity in subjects with relapsed or refractory NHL.
