ASC-201 (Oncoral) is an investigational oral formulation of irinotecan, a topoisomerase I inhibitor, being developed by Ascelia Pharma. Irinotecan is a camptothecin derivative that inhibits DNA topoisomerase I, leading to double-strand DNA breaks and cell death. While irinotecan is traditionally administered intravenously, ASC-201 is designed as a small-molecule oral tablet to improve patient convenience and potentially offer a better safety profile through different dosing schedules. It is primarily being evaluated for the treatment of advanced gastric cancer, particularly in the third-line setting, often in combination with other agents like trifluridine/tipiracil (Lonsurf).
