Overview

ASC-41 is a once-daily oral, liver-targeted small molecule prodrug developed for the treatment of non-alcoholic steatohepatitis (NASH). After administration, it is metabolized in the liver by CYP3A4 to its active form, ASC41-A, which acts as a potent and selective agonist of thyroid hormone receptor beta (THRβ). Activation of THRβ in the liver regulates lipid metabolism and reduces hepatic fat accumulation. Clinical studies have shown that ASC-41 significantly reduces low-density lipoprotein cholesterol (LDL-C) and triglycerides. The drug is being developed by Ascletis and Gannex Pharma, with ongoing phase II clinical trials for NASH. It has also been studied in overweight/obese subjects and patients with non-alcoholic fatty liver disease (NAFLD)[1][2][3][4][5][8].
