Overview

ASC101 is an investigational chimeric antigen receptor natural killer (CAR-NK) cell therapy developed by Ascle Therapeutics. Built on the NK-92 cell platform, ASC101 is engineered to target Roundabout homolog 1 (ROBO1), a cell surface receptor that is highly expressed in approximately 80% of solid tumors, including breast cancer, but has limited expression in normal tissues. The mechanism of action involves the recognition of ROBO1-expressing tumor cells by the CAR-NK cells, which then mediate direct tumor cell killing through NK cell-directed cytotoxicity. ASC101 is currently in the IND-enabling and IND application stage, with preliminary data from investigator-initiated trials (IIT) showing a disease control rate of 76.5% in patients with solid tumors.
