Overview

ASC30 is an investigational small molecule GLP-1 receptor (GLP-1R) biased agonist developed by Ascletis Pharma for the treatment of obesity. It is unique in that it can be administered both as a once-daily oral tablet and as a once-monthly subcutaneous injection, offering significant flexibility in dosing regimens. ASC30 demonstrates two- to threefold greater in vitro potency against GLP-1R compared to orforglipron, another small molecule GLP-1R agonist, and has shown superior pharmacokinetic properties including a long half-life (up to 60 hours orally and 36 days subcutaneously) and high oral bioavailability. In clinical studies, ASC30 has produced substantial weight loss (up to 6.3% mean body weight reduction over four weeks) with a favorable safety profile—most adverse events were mild or moderate gastrointestinal effects, with no serious adverse events reported. The drug does not recruit β-arrestin, which may contribute to its differentiated efficacy and safety profile[1][2][5][6][7][8].
