ASC39 is an orally bioavailable, potent, and selective small molecule amylin receptor agonist developed by Ascletis Pharma for the treatment of metabolic diseases, primarily obesity. Discovered using the company's Artificial Intelligence-assisted Structure-Based Drug Discovery (AISBDD) technology, ASC39 features a unique chemical scaffold. It demonstrates high potency for the human amylin 1 receptor (hAMY1R) with an EC50 of 21.4 pM and shows high selectivity over the calcitonin receptor (hCTR). In preclinical diet-induced obese rat models, ASC39 has shown significant weight loss efficacy comparable to eloralintide. It is being developed as both a monotherapy and for use in combination with Ascletis' GLP-1 receptor agonist, ASC30.
