ASC47 + semaglutide is an investigational **combination therapy** for obesity that pairs ASC47, an adipose-targeted, ultra-long-acting subcutaneous small molecule **thyroid hormone receptor beta (THRβ) selective agonist**, with **semaglutide**, a Glucagon-like peptide 1 receptor (GLP-1R) agonist. Both act via different, complementary mechanisms: ASC47 targets adipose tissue to promote weight loss while preserving muscle mass, whereas semaglutide reduces appetite and food intake. Preclinical studies show the combination produces **superior weight loss and greater muscle preservation** compared to semaglutide alone, with a 56.7% greater reduction in body weight in diet-induced obese mouse models. Human Phase 1 trials (ASC47-103, NCT06972992) are ongoing to evaluate safety and preliminary efficacy in obese adults without type 2 diabetes. ASC47 is developed in-house by Ascletis and administered as a once-monthly subcutaneous injection, while semaglutide is given weekly. This is the first known clinical investigation combining a THRβ-targeted drug with an incretin for obesity therapy[1][2][3][4][6][7][8].
